A marine bacterial adhesion microplate test using the DAPI fluorescent dye: a new method to screen antifouling agents ArchiMer

Aims: To develop a method to screen antifouling agents against marine bacterial adhesion as a sensitive, rapid, quantitative microplate fluorescent test. Methods and Results: Our experimental method is based on a natural biofilm formed by mono-incubation of the marine bacterium Pseudoalteromonas sp. D41 in sterile natural sea water in a 96 wells polystyrene microplate. The 4'6-diamidino-2-phenylindole (DAPI) dye was used to quantify adhered bacteria in each well. Total measured fluorescence in wells was correlated with the amount of bacteria showing a detection limit of one bacterium per 5 µm2 and quantifying 2 x 107 to 2 x 108 bacteria adhered per cm2. Action de l'acétate de tributyle-étain sur les oeufs et les larves D de deux mollusques d'intérêt commercial : Crassostrea gigas (Thunberg) et Mytilus galloprovincialis (Lmk) ArchiMer

The Tributyl tin acetate, a component of certain antifouling paints, has deleterious effects on eggs and D shaped larvae from Crassostrea gigas at 5 p.p.b. (HIS & ROBERT, 1980). Here we have tested concentrations as low as 1 p.p.b. on eggs and on 24 hours old larvae both from Crassostrea gigas and Mytilus galloprovincialis. The same results are got ; this product kills embryos and larvae in a few days ; it prevents growth of 24 hours old larvae exposed to this toxic. Mussels larvae seem to be less sensitive.

Within this study, the potentiality of DBHB against microfouling has been investigated. Crude polysaccharides from the red algae Corallina officinalis (SP1) and Pterocladia capillacea (SP2) were extracted and characterized according to their chemical content and their antioxidant, anti-inflammatory, anticoagulant, antibacterial, antifungal, and antifouling activities. European Community regulations on chemicals promote alternative methods to test substances presenting potential risks for the environment. In the present work, cultured atrial cells isolated from oyster (Crassostrea gigas) were used as an experimental model to investigate the toxicity of tributyltin (TBT) after short-time exposure at concentrations representative of those that can be measured in seawater, marine sediments and/or bivalves bioaccumulating this pollutant. The antifouling potential of commercial hydrolases, four proteases, seven glycosidases and one lipase was evaluated on the adhesion of marine Pseudoalteromonas sp. D41. The experimental method, adapted to screen antifouling agents, was based on bacterial adhesion in natural sterile sea water in a microtiter plate and on total biomass quantification by the fluorescent dye DAPI (4[prime]6-diamidino-2-phenylindole). Aims: The nature of exopolymers involved in the adhesion of a marine biofilm-forming bacterium Pseudoalteromonas sp. D41 was investigated to evaluate and understand the antifouling potential of subtilisin. Methods and Results: The exopolymers of D41 produced by fermentation were analysed by FTIR and SDS-PAGE showing the presence of polysaccharides, glycoproteins and proteins. A high content of proteins was detected both in soluble and capsular fractions. A wild strain of Chaetoceros calcitrans and wild and diuron-resistant strains of Tetraselmis suecica, were exposed to the PSII inhibitor herbicides diuron and irgarol, individually and in mixtures. The effects of three concentrations of diuron and irgarol and four binary mixtures were evaluated on doubling time, relative reactive oxygen species and lipid content by flow cytometry, and on photosynthetic efficiency by pulse amplitude modulated fluorescence. Four new brominated tyrosine metabolites, aplyzanzines C–F (1–4), were isolated from the French Polynesian sponge Pseudoceratina n. sp., along with the two known 2-aminoimidazolic derivatives, purealidin A (5) and 6, previously isolated, respectively, from the sponges Psammaplysilla purpurea and Verongula sp. Their structures were assigned based on the interpretation of their NMR and HRMS data. The compounds exhibited quorum sensing inhibition (QSi) and antifouling activities against several strains of bacteria and microalgae. To our knowledge, the QSi activity of this type of bromotyrosine metabolite is described here for the first time.
